This study was performed to explore the protective and curative effects of safranal against the toxicity of an anticancer drug, tamoxifen especially hepato-nephrotoxicity. Therefore, 30 adult female were allocated into 5 equal groups and IP injected with normal saline (Control), 0.05mg safranal /kg b.wt for 9 days (Saf), 45mg tamoxifen/kg b.wt for 6 days (TAM), 0.05mg safranal/kg b.wt for 9 days before tamoxifen intoxication (Prot), and 0.05mg/kg b.wt of safranal for 9 days after tamoxifen intoxication (Cur). Tamoxifen injection induced oxidative stress, which confirmed by a significant decrease (p <0.05) of the activities catalase, superoxide dismutase, glutathione peroxidase and glutathione reductase, and elevation of activity of glutathione S- transferase and malondialdehyde concentration. Moreover, it provoked a significant increase (p <0.05) of alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, urea and creatinine, which indicated its hepato-nephrotoxic effects. Interaperitoneal injection of safranal either as protective or curative could ameliorate these toxic effects of tamoxifen. Therefore, it could be concluded that safranal administration has protective and curative effects against hepato-nephrotoxicity which induced by tamoxifen in female rats.
